Hybrid Antibodies with Dual Specificity for the Delivery of Ricin to Immunoglobulin-bearing Target Cells1

چکیده
Hybrid antibodies possessing one binding site for the toxic lectin ricin and a companion site directed against human immunoglobulm were constructed in vitro. This bifunctional re agent specifically attached to human lymphocyte surface immunoglobulin determinants and, thus situated, could simulta neously capture ricin molecules or its toxic A chain. Attachment of these components to the cell was revealed by specific fluorescein-labeled antibodies. Once concentrated at the target cell membrane, hybrid-bound toxin was subsequently released to function via its normal mechanism of biological action. It gained access to ribosomes, its intracellular target, and cur tailed protein synthesis. Toxicity was not augmented for immunoglobulin-negative cells to which hybrid could not bind and free human immunoglobulin G could competitively block the enhanced effects observed for immunoglobulin-bearing cell lines. These results indicate that hybrid antibodies may be utilized to carry active agents within close proximity to the membrane of a specified cell type and thereby selectively enhance their effect.

متن کاملKinetics of protein synthesis inactivation by ricin-anti-Thy 1.1 monoclonal antibody hybrids. Role of the ricin B subunit demonstrated by reconstitution.
Antibody-toxin conjugates of different compositions have been reported to have varying differential toxicities between target and non-target cells in uitro. In this report, we compare the kinetics of protein synthesis inhibition of several different types of anti-Thy 1.1-ricin hybrids. Utilizing two monoclonal antibodies which vary in affinity for the Thy 1.1 antigen by >IO3, thioether-linked ...

Objective(s): CD19 is a transmembrane glycoprotein of immunoglobulin superfamily. In order to treat lymphoma, monoclonal antibodies (mAb) can target different antigens, including CD19, CD20 and CD22 on the surface of B-cells. Along with biotechnology progress, a new generation of antibodies is introduced, with the purpose of eliminating the defects of the previous generation. Among the most dev...

Recently it has been reported, that immunoglobulin Y (IgY) can be used instead of polyclonal antibodies extracted from mammals (IgG) for the purpose of diagnosis and therapy. These antibodies are found to have better properties in terms of specificity and ease of large-scale production. In addition, IgY binds neither to mammalian complement or Fc-receptors nor does it interfere with rheumatoid ...

Hybrid, bivalent antibody molecules bearing specific combining sites for both ovalbumin and bovine gamma globulin were produced by reoxidation of a mixture of the 3.5S fragments of the two specifically purified antibodies. The dual specificity and the properties of the hybrid antibody were demonstrated by mixed agglutination and two stage agglutination experiments, and by test systems utilizing...
